Abstract
"幾丁聚醣會明顯降低血脂、血糖,過去幾年有關幾丁聚醣降低血糖、血脂的 機制探討,我們利用糖尿病動物及肥胖動物已解開其機制,在過程中,我們發現 幾丁聚醣會明顯降低糖尿病及肥胖大鼠肝臟脂質,顯示幾丁聚醣對脂肪肝具有改 善的效果,值得注意的是我們發現以血脂的下降及糞便脂質的排出增加並無法完 全解釋其機制。另一方面糖尿病大鼠餵食幾丁聚醣4週,發現幾丁聚醣具有降血 脂、肝脂的效果,但9週後降血脂效果消失,而肝脂質的降低效果卻仍明顯存在。 由於血脂的變化與肝脂代謝關係密切,此結果顯示長期或短期攝食幾丁聚醣對肝 臟脂質代謝可能不一樣。此外許多研究報告指出高分子量幾丁聚醣與低分子量幾 丁聚醣對血脂及肝脂的降低效果不一樣,因此為瞭解幾丁聚醣對肝臟脂質代謝影 響的機制,我們計畫作三年的研究,期待釐清幾丁聚醣對脂肪肝影響之機制。(1) 第一年: 幾丁聚醣改善非酒精性脂肪肝的機制探討。(2) 第二年:探討長期(10 週)、短期 (4 週) 餵食幾丁聚醣對糖尿病動物肝臟脂質代謝的影響。(3) 第三 年: 比較高、低分子量幾丁聚醣對肥胖動物脂肪肝影響機制之探討。" "Recently, we focused on the investigation the mechanism of reducing plasma glucose and lipids by chitosan. We found that chitosan decreases plasma lipids through increasing bile acid excretion, fecal lipids and intestinal short chain fatty acids production. Lower plasma glucose may be due to the reduced hepatic gluconeogensis and intestinal disaccardiase activities after chitosan treatment. It is worth to note that reduced hepatic lipids were also observed in obesity and diabetic rats, indicating that chitosan may ameliorate the fatty liver. However, it is hard to explain why the hepatic lipids were reduced through the increased fecal lipids and decreased plasma lipids. On the other hand, long-term feeding of chitosan can reduce hepatic lipids but not plasma lipids in DM animal model. The plasma lipids reducing effect only observed in short-term study. The mechanism is still unknown. In addition, a comparative effect of high molecular weight and low molecular weight chitosan on plasma lipids and hepatic lipids is also controversy. In order to investigate the mechanism of ameliorating fatty liver by chitosan, the project including 3-years plan will be studied. 1st year: Study on the effect of chitosan on non-alcohol fatty liver. 2nd year: Effect of long-term or short-term chitosan feeding on plasma lipids and hepatic lipids in type 2 diabetic rats. 3rd year: A comparative effect of high molecular weight and low molecular weight chitosan on fatty liver in obesity animals. "
