Description

Abstract
石花菜是台灣盛產的海藻,大部分產於北部與東北角海岸,屬於安曼司石花菜(Gelidium amansii)。石花菜含有許多水溶性纖維,在日本被稱為寒天,已有超過 350 年的食用歷史。我們先前的報告指出安曼司石花菜可以顯著降低禁食血液胰島素濃度、總膽固醇濃度、三酸甘油酯濃度及胰島素抗性,顯示省產石花菜有助於調節脂質代謝與糖代謝。進一步將關華豆膠(guar gum)與安曼司石花菜水萃物作比較,結果發現安曼司石花菜水萃物在降血脂功能方面與關華豆膠幾乎不相上下,且具有更有高的脂解能力。我們推測安曼司石花菜調節脂質代謝與糖代謝的生理功能可能與腸道生理有密切關係。1.調節腸道生理代謝方面:推測安曼司石花菜會降低SGLT1,也會增加Angptl4在腸道的表現量,降低胰脂解酶的活性,影響FATP4及FABP2的表現並進一步影響腸道GLUT2蛋白表現。2.調節脂質代謝:我們已證實石花菜的熱水萃取物可能會影響SREBP1-C及SREBP2 的表現,進而影響PPAR-γ。因此我們推測石花菜的熱水萃取物可能會抑制肝臟ApoB,增加MTTP蛋白質表現,減少肝臟脂肪的堆積。因此我們計畫利用大鼠作成高脂及糖尿病動物模式來探討石花菜的熱水萃取物對脂質代謝與糖代謝影響的機制,期待釐清石花菜的熱水萃取物對影響血脂與血糖之機制。 Gelidium amansii (GA) is an edible red algae that is distributed mainly in northeastern Taiwan. GA is rich in soluble dietary fiber. Its agar product is known as Kanten in Japan, has more than 350 years of edible history. We have reported that GA can decrease plasma triglyceride, total cholesterol and insulin levels and insulin resistance. However, the mechanism is still unclear. In addition, A further comparison between guar gum and GA hot water extracts (GAE) on the hypolipidemic and hypoglycemic actions showed that they are almost comparable. We speculate that the physiological function of GAE regulation of lipid metabolism and glucose metabolism may be closely related to intestinal physiology. We also speculate that 1. GAE may decrease intestinal SGLT1 and increase Angptl4 expression to reduce lipase, then affect GLUT2 expression. 2. GAE may inhibit hepatic ApoB and increase MTTP protein expressions to reduce hepatic lipid accumulation. The aim of this study is to investigate the beneficial effect and the possible molecular mechanism of GAE on lipid and in high-fat-diet-fed rats and diabetic rats.
