Description

Abstract
異位性皮膚炎是一種好發於孩童的慢性發炎性皮膚病,臨床症狀可見皮膚癢和皮膚乾燥。全球大 約有5-20 %的孩童受此困擾。目前對異位性皮膚炎的治療主要以類固醇為主,雖然可有效控制皮膚炎病情,但長期使用會有造成皮膚萎縮、血管擴張及影響腎上腺素分泌而影響生長等副作用,造成很多病人及家屬會尋求健康食品等另類替代治療。近年來的研究文獻指出海藻硫酸多醣具有抗病毒、抗發炎、抗腫瘤、抗血栓、提高免疫力等保健功效。本計畫主要利用細胞實驗及小鼠動物模式探討水雲 (Cladosiphon okamuranus) 硫酸多醣萃取物對於改善異位性皮膚炎及免疫調節的效果。這是一個延續性計畫,目前核定的一年計畫中已進行水雲多醣萃取物處理P815肥大細胞實驗,以細胞存活率(MTS assay)、β-胺基己醣苷梅脫顆粒測定(β-hexosaminidase degranulation assay) 及細胞因子試驗含量變化等實驗進行異位性皮膚炎改善及調節免疫效果評估,進一步分析水雲多醣萃取物改善異位性皮膚炎及免疫調節之機制。在未來申請的一年計劃中,將利用DNCB誘導異位性皮膚炎的BALB/c小鼠模式,探討口服水雲多醣與塗抹水雲多醣萃取物乳膏改善異位性皮膚炎及免疫調節的效果及其作用機制。延伸至模擬異位性皮膚炎療程實驗,將水雲多醣萃取物乳膏作為保濕劑聯合治療之使用,檢視其患部恢復之成效與是否具有降低類固醇藥物副作用之效果。最後,以內服外用方式,使用小鼠異位性皮膚炎模式測試水雲多醣和水雲多醣萃取物乳膏,對於異位性皮膚炎時所引發過敏反應,是否具有加乘免疫調節之效果。經由上述的研究,我們希望能發展出有效的水雲多醣萃取物產品類型,以作為改善異位性皮膚炎及免疫調節健康食品之應用。 Atopic dermatitis (AD) is a chronic inflammatory skin disorder that occurs most frequently in children. AD is characterized by itchy and dry skin and it affects approximately 5 to 20% of children in the world. Currently, the treatment that is most widely used to control AD is corticosteroids. Corticosteroids is effective in treating AD, however, there are concerns about the side effects such as skin atrophy, vasodilation, and inhibits the growth of children by affecting their adrenal gland function. Therefore, there are many patients and families who seek complementary and alternative treatments such as health food. Recently, the sulfated polysaccharide extracts of seaweed have been shown broad biological activities, including antiviral, antitumor, immunomodulatory and anticoagulant activity. This study will investigate the sulfated polysaccharide extracts of Cladosiphon okamuranus for improve the atopic dermatitis and immunomodulation on in vitro and in vivo model. This is a continuity plan and in the currently approved one-year plan, we use cell viability assay (MTS assay) to evaluate the toxicity of the sulfated polysaccharide extracts of Cladosiphon okamuranus. We will utilize β-hexosaminidase degranulation assay and cytokine expression (ELISA) to study the protective effects of the sulfated polysaccharide extracts of Cladosiphon okamuranus. In the next year proposal, we will investigate the improvement effects and immunomodulation mechanisms of atopic dermatitis treated orally and topically with the sulfated polysaccharide extracts of Cladosiphon okamuranus on the DNCB induced BALB/c mice model. Furthermore, we also combined corticosteroids and the cream of the sulfated polysaccharide of Cladosiphon okamuranus treated the DNCB induced BALB/c mice which attempt to improve the side effect of corticosteroids. Finally, we combined oral and topical the sulfated polysaccharide of Cladosiphon okamuranus for the treatment of the DNCB induced BALB/c mice. Through all these efforts, we hope to develop that sulfated polysaccharide extracts of Cladosiphon okamuranus may be used as the health food supplement to improve the atopic dermatitis and immunomodulation.
