Description

Abstract
蛋白質經酵素水解後, 其水解產物中之短鏈胜肽 具有抑制血管升壓素轉換酶 活性的作用, 而可降低血壓.因此, 本計畫的研究目的, 擬以本省鯵 魚為原料, 利用不同蛋白酶 進行魚肉蛋白質的水解.水解產物以高效率液體層析法進行劃分後, 探討各劃分物對血管升壓素轉換酶 的抑制效果及此胜肽 的純化與鑑定.並將水解產物餵食原發性高血壓老鼠後, 測定其血壓的變化情形, 以了解水解產物對原發性高血壓老鼠降低高血壓的生理效果.在本研究中擬探討的工作項目包括: ( 1 )酵素的水解.( 2 )胜肽 的劃分.( 3 )胜肽 對血管升壓素轉換酶 的抑制與降血壓的效果.( 4 )胜肽 的純化與定序分析.因此, 由本研究中確定鯵 魚肉水解物中具抑制ACE活性之機能性物質的組成與氨基酸序列, 及了解鯵 魚肉水解物中寡胜肽 降高血壓的保健效果. The hydrolysates contain short chain peptides which having the inhibitory activities against Angiotensin I-converting enzyme when the protein was hydrolyzed, and could reduce the blood pressure.The purpose of this study will be to use the horse mackerel as a material, and undertake fish protein hydrolysis using various protease, to produce hydrolysates.The hydrolysates were fractionated by high performance liquid chromatography, and to investigate the inhibitory effects of different fractionated products on Angiotensin I converting enzyme.The major peptides were further purified and identified by reverse-phase HPLC.Also, the blood pressure will be determined after the hydrolysates was fed to spontaneously hypertensive rat, and to understand the physio-logical effects of hydrolysates reducing on blood pressure of spontaneously hypertensive rat.The research items include: ( 1 )protein hydrolysis, ( 2 )fractionation of peptides, ( 3 )inhibitory effect on angiotensin I converting enzyme and anti-hypertensive effect, and ( 4 )purification and identification of peptides.From this study, the bioactive peptides inhibitory effect on angiotensin I converting enzyme and physiological effect on anti-hypertension will be established.
