Description

Abstract
"本研究室自環境中成功獲得一株具血糖調節活性之酵母菌,其活性物質稱 為葡萄糖耐量因子 (GTF)。利用所建立的細胞培養模式分析GTF 活性,建立該 株菌生產GTF 之培養基及培養條件。然而由於酵母菌細胞壁特別堅固,人類無 法分解,未來實際應用於人體時可能將因菌體無法被消化而致無法顯示其活性。 本研究室雖已建立氨水萃取菌體GTF 活性物質,以細胞模式完成其作用機轉探 討。然而未來應用時由於菌體需經萃取、離心、減壓濃縮(去除溶劑)及凍乾等操 作,操作煩雜不易量產。合作廠商極希望以破碎菌體方式代替萃取,以利未來量 化製程操作,因此,在人體應用之前,需先確定該菌體、GTF 活性萃取物、以 及破碎菌體對糖尿病大鼠之降血糖效果。本研究首先將分別製備完整酵母菌體、 酵母菌GTF 萃取物及破碎酵母菌體三種樣品,接著以口服方式,探討這三種樣 品對STZ 誘導之第一型糖尿病大鼠及高果糖誘導之第二型糖尿病大鼠之葡萄糖 耐受性及血糖、血中胰島素及尿糖之影響。期望藉由本研究確認三種樣品未來在 兩種不同類型糖尿病之應用潛力。同時,期望以破碎酵母菌體取代萃取方式,未 來可簡化製程,提高產率。" "A specific yeast strain with hypoglycemic activity (called GTF activity) has been isolated from soils and investigated. The cultural medium and conditions for GTF production by this yeast strain have been established using a cell-line model for GTF activity assay. GTF is present inside cells, and human can not digest the tough cell wall of yeast. Therefore, little hypoglycemic activity of this yeast strain may be obtained due to being indigestible by human if it is directly ingested. The protocol for GTF extraction from yeast cells has been eatablished. However, the tedious procedures including extration, centrifugation, vaccum concentration and freeze-drying cause the difficulty in industrial production. The cooperative company urgently wants to know the possibility for future application by physically breaking the yeast cells. Therefore, three samples including whole yeast cells, GTF extract, and pressure-breakage yeast cells will be prepared first in this project. The hypoglycemic effects and glucose tolerance activities of these 3 samples on the STZ-induced type I diabete rats and high-fructose-induced type II diabete rats will be investigated by oral feeding thses samples."
