Abstract
"目前人工栽培靈芝以赤芝 (Ganoderma lucidum) 與松杉靈芝 (G. tsugae)為主。多醣是靈芝子實體主要生理活性主物質之一,具有多種生理 活性。由於靈芝子實體栽培耗時費力與空間,故液態培養靈芝漸受重視。 液態培養菌絲體多醣與胞外多醣具有抗腫瘤、免疫調節及護肝功效等多種 活性,其活性與多醣分子量、分子結構與水溶性有關。 培養基組成與培養條件明顯影響靈芝菌絲與胞外多醣的產量與活 性,本研究室已建立液態培養靈芝之最佳培養基配方與培養條件。本計畫 首先欲建立靈芝複合菌株之發酵條件,以增加菌絲產量及得到不同分子量 分佈之胞外多醣;接著分離純化胞外多醣,分析其分子量及免疫調節活 性。本計畫首先將選取8 株靈芝菌株,以所建立配方培養基(Gano medium) 及條件先分別培養各單一菌株,從中選取生長最好的 5 株菌。接著進行 多重複合菌株發酵,篩選出菌絲體與胞外多醣產量最高之組合菌株,建立 其發酵條件。最後更分離純化組合菌株之胞外多醣,分析其分子量;並以 小鼠巨噬細胞株 RAW 264.7 之體外分析模式,探討不同分子量胞外多醣 對細胞增生、細胞型態、吞噬活性、超氧陰離子及NO 分泌、細胞生命週 期及細胞表現類樹狀細胞表面抗原之影響,以初步了解其免疫活性。" "Ganoderma lucidum and G. tsugae are the main cultivated Ganoderma species. The polysaccharides are one of the main bioactive components for Ganoderma species. Although the polysaccharides from fruiting body of the cultivated Ganoderma species have various physiological activities, it is time- and space-consuming, and labor-intensive for cultivation of the fruiting body of Ganoderma species. Recently, people are more focus on liquid cultivation of Ganoderma species. The bioactivities of mycelium and extracellular polysaccharides (EPS), including anticancer, immune modulation and protection from liver damage, have been demonstrated. The molecular size and structure, and solution property of these polysaccharides can affect their bioactivities. The optimized medium composition (Gano medium) and cultural conditions have been established in this laboratory for cultivation of single strain of Ganoderma lucidum. The aims of this project are to establish the co-cultivation conditions for multiple strains of Ganoderma species to increase the mycelium production and EPS size distribution, and to purification and investigation of the molecular sizes and immune modulation activity of various EPS. Total 8 species of G. lucidum and G. tsugae will be cultivated individually in Gano medium; among which 5 species with higher mycelium production will be screened out for the second stage test of multiple strain cultivation. Accordingly, the optimal condition for co-cultivation of multiple strains of Ganoderma species will be established. Finally, various molecular sizes of EPS in the culture filtrate will be purified. Their immune modulation activities will be investigated in vitro using the cell line of a murine macrophage RAW 264.7, based on their effects on cell morphology, cell proliferation, cell cycle arrest, dendritic surface antigen distribution in cells, phagocytic activity, superoxide anion and nitric oxide secretion."
