Description

Abstract
"北蟲草 (Cordyceps militaris)具抗菌、抗發炎、降膽固醇、抗癌、及增強免 疫力等生理活性。其活性成分主要以蟲草素、腺苷、蟲草酸及蟲草多醣最受重視。 固態培養北蟲草菌絲體所得蟲草素與腺苷含量高於液態培養所得。 憂鬱症已成為二十一世紀高負擔三大疾病之一, WHO 預估於 2030 年 其將成為世界疾病負擔排名第一。由於目前臨床抗憂鬱藥物常有副作用,許多研 究指出γ-胺基丁酸(γ-aminobutyric acid,GABA)具抗憂鬱活性。本研究室已從上百 株分離自魚腸道之乳酸菌分離株篩選出GABA產量最高的一株乳酸菌FPA3709, 經鑑定其為 Lactobacillus brevis。在前置試驗中,取商業上割除北蟲草子實體後 的剩餘米基為原料,以FPA3709進行液態培養之初步研究,得到極高GABA 含量 (17 mg/mL)之發酵液,其GABA量高於許多已發表的文獻。本計畫欲進行北蟲草 與FPA3709乳酸菌(產GABA 乳酸菌) 二階段發酵培養之研究,以生產高GABA 與蟲草素含量之抗憂鬱活性產品。第一年將建立北蟲草與乳酸菌共同發酵培養條 件。將採用固態培養方式培養北蟲草菌絲體,然後加入無菌水進行乳酸菌深層靜 置發酵。由於發酵產品未來可直接置於適當溫度下販售,或者經適當乾燥方法處 理得到粉末狀產品。第二年研究首先探討不同加熱條件對發酵液中乳酸菌及各活 性成份含量之影響,進而提供噴霧乾燥操作條件參考。接著探討噴霧乾燥與冷凍 乾燥法所得發酵粉末中活性成分含量差異。最後探討不同儲存溫度對發酵液及乾 燥粉末中活性成分含量之變化。第三年採用臨床抗憂鬱藥物篩選常用的強迫游泳 試驗動物模式,並以目前常用抗憂鬱藥物『百憂解』為正對照組,探討發酵液及 乾燥粉末產品之抗憂鬱活性及其可能機轉。 " "Cordyceps militaris has various physiological activities including antimicrobial, anti-inflammatory, hypochlosterol, anti-cancer and immune-enhancing activities. The active components in C. militaris include cordycepin , adenosine, cordycepic acid and polysaccharides. Higher amounts of mycelium, cordycepin and adenosine are obtained by solid state cultivation of C. militaris, compared with submerged cultivation of this species. Depression is a widespread incapacitating psychiatric ailment ranked by the World Health Organization as one of the most burdensome diseases of society. Despite the prevalence and severe effects of this disease, the efficacy of currently available antidepressants is often inconsistent, and many of them exert undesirable side effects. Several studies in recent years have demonstrated the antipressant activity of GABA. One lactic acid bacterium (LAB) FPA3709 with high GABA production potential was chosen from hundreds of LABs isolated from fish intestines and was further identified as Lactobacillus brevis. C. militaris is widely cultivated using cooked rice as the basal solid medium. In a preliminary study FPA3709 produced a very high amount of GABA(17 mg/mL) in the suspension of rice residues from the fruiting body of C. militaris. In this project a two- stage incubation of C. militaris and FPA3709 will be established for the production of a GABA- and cordycepin-rich novel product with antidepressant activity. In the first year, the protocol for the 2 stages of incubation of C. militaris and FPA3709 will be established. Koji by incubation of C. militaris in rice will be prepared. After adding water to C. militaris koji, FPA 3709 will be inoculated and incubated. In 2nd year, effects of heating on the amounts of individual active components in the fermented product will be investigated. Then, the novel ferment will be spray-dried and freeze-dried. The amounts of the active components in dried products using two-types of drying methods will be compared. Finally, the changes in the active components of fermented products (liquid, and dried powder) during storage at different temperatures will be investigated. In the third year, the antidepressant effects of the fermented products will be investigated using forced swimming rat model. The underlying mechanism for the antidepressant effect of GABA will also be investigated. "
