Description

Abstract
"靈芝具有抗腫瘤、免疫調節、護肝、抗氧化、降血糖與抗老化等多種生理活性, 其活性成分包括多醣類、三萜類 (triterpenoids), superoxide dismutase (SOD) 與類固醇 (steroid)。目前人工培養靈芝主要為固態子實體培養與液態菌絲體培養。固態子實體培 養時間長、成本高;液態培養靈芝菌絲體培養時間雖較短,因含大量水分,所得活性 物質量相對低很多。若將其乾燥成粉末,耗費許多能源。近年來發展固態發酵生產靈 芝菌絲體,與固態培養子實體相比,大幅縮短培養時間;與液態菌絲體相比,可增加 單位體積菌絲體產量及提高菌絲體三萜類含量。γ-胺基丁酸(γ-aminobutyric acid, GABA) 為人體自然產生具抑制神經傳導功能的非蛋白質胺基酸,有降血壓、改善糖尿病、活 化肝腎、抗憂鬱、改善癲癇、帕金森氏和老年癡呆症等活性。氧化壓力對細胞的損傷 已被認為是造成老化及許多神經退化相關疾病的最原始原因,乙型澱粉胜肽堆積會造 成神經細胞凋亡。本研究室已建立靜置式固態培養靈芝菌絲培養之培養基配方,發現 靈芝菌絲生長時可產生GABA。本計畫第一年將以新穎固態發酵槽培養靈芝菌絲體,探 討溫度、攪拌及通氣對靈芝菌絲體生長及生產 GABA 之影響,並分析靈芝發酵產物之 活性成分與抗氧化活性。第二年將以神經細胞株分析模式,探討含 GABA 之靈芝產物 對氧化壓力及乙型澱粉胜肽傷害神經細胞的保護效果。除了確認含GABA固態靈芝發酵 物未來應用在神經功能異常疾病之防治,也希望能釐清其真正功效成分為何。" Ganoderma lucidum containing active components of polysaccharides, triterpenoids, superoxide dismutase and steroids has various bioactivities including anticancer, immune modulation, liver protection, antioxidant, hypoglycemic and antiaging. The solid-state fermentation (SSF) of fruiding body and submerged cultivation of mycelium are the two widely used methods for cultivation of G. lucidum. It takes long time for fruiting body by SSF; while the bioactivity will be lower due to the presence of large amount of water for submerged mycelium cultivation and drying mycelium culture is high energy cost. Recently, an alternative mycelium SSF is provided for cultivation of G. lucidum. This method is quicker, compared to SSF fruiting body, and higher amounts of mycelium and triterpenoids are produced by SSF mycelium method, compared to submerged mycelium method. y-Aminobutyric acid (GABA) is a non-protein amino acid and a inhibitor of neuron transmittance in human with bioactivities of hypopressure, improvement of diabetic symptom, activating liver/kidney function, anti-depression, improvement of epilepsy, Parkinson and Alzheimer’s diseases. The neuron damage by oxidative stress will initialize the aging and neural degeneration related deseases. The (3- amyloid peptide aggregation will cause the apoptosis of nerve cells. In this laboratory the cultivation medium has been formulated for the production of mycelium and GABA by G lucidum using SSF. In the 1st year of this project the effects of cultural conditions of agitation, aeration and temperature on the production of mycelium and GABA by G lucidum using a novel solid-state fermentor will be investigated. The active components and the antioxidant activity of the fermented product will be also analyzed. In the 2nd year the protective effects of the GABA-containing fermented product on oxidative stress- or 3-amyloid peptide- induced neuron toxicity will be evaluated.
