Description

Abstract
利用自行設計適用於河魨魚種之引子組 (L newset-1 & H 15149),增 幅隨機採樣之台灣市售疑似河魨加工品 46 件檢體中 376 bp 之 cytochrome b 基因序列,合併利用限制酶 Bsa JI,Hinf I,Sap I,Taq I 及 Aci I 之片段長度多型性,鑑別檢體中,宜蘭及台北各一件魚柳為不 明魚種,台北一香魚片檢體為瀧紋河魨,台南及高雄各有一香魚片檢 體為毒鯖河魨,台北及台南各有一香魚片檢體、高雄香魚片二檢體為 白鯖河魨,其餘香魚片之原料魚種皆為黑鯖河魨。另利用此引子組 (L newset-1 & H 15149),增幅疾病管制局抽檢香港進口冷凍魚排 8 件檢體 中 376 bp 之 cytochrome b 基因序列,合併利用限制酶 Bsa JI 之片段長 度多型性,鑑別 8 件檢體其原料魚種皆為黑鯖河魨。其次利用之引子 組 (L newset-1 & H 15149),增幅自大陸舟山市漁獲市場挑選疑似以有 毒河魨製造之留尾香魚片 6 件檢體中 376 bp 之 cytochrome b 基因序 列,合併利用限制酶 Bsa JI,Hinf I 及 Sap I 之片段長度多型性,鑑別 6 件檢體其原料魚種均為毒鯖河魨。To understand the application of DNA techniques in identifying species of puffer fish products, 60 samples of fresh meat of lunch box, dried dressed fish fillets, and roasting dried dressed fish fillets were collected from different cities such as Yilan, Keelung, Taipei, Hsinchu, Tainan, Kaohsiung, Pingtung, Hongkong, and Zhoushanshi in Mainland China and tested. It was found that one sample from Taipei was T. oblongus, each one sample from Tainan and Kaohsiung was L. lunaris, 6 samples of dried dressed fish fillets purchased from Zhoushanshi in Mainland China all were L. lunaris, each one sample from Taipei and Tainan was L. wheeleri, two samples from Kaohsiung were L. wheeleri, each two samples of roasting dried dressed fish fillets from Yilan and Taipei were un-identified, and all of other samples were L. gloveri. These results indicated how to control puffer products is a serious problem for the government.
