"在台灣，海洋生物毒菟葵毒、河魨毒和麻痺性貝毒引起之食物中毒事件屢次發生，且發生死亡事件，甚為國人所重視，申請者以往進行了一系列河魨毒和麻痺性貝毒之毒性探討，建立了相當完整的魚貝類毒性和魚貝類基因資訊。2011 年，台灣東部花蓮地區發生食用小型鯡魚之死亡事件。該魚種經本研究利用基因鑑定係屬鯡魚 Clupea pallasii，毒成分經純化，以 LC-MS/MS 鑑定確定為菟葵毒。菟葵毒易形成衍生物，很難純化鑑定，其中毒症狀為肌肉溶解症、心肌麻痺、肌酐酸磷酸酶活性急劇上升，人中毒易引起死亡事件，但鼠類試驗之生物毒性相當難。台灣以前曾發生箱魨魚、翻車魚、鯡魚、臭都魚等是疑似含有菟葵毒之危害問題。為繼續上述成果，本計畫第 1 年擬利用研究加以建立之溶血試驗法、肌酐酸磷酸酶活性測定法配合小白鼠之生物毒性試驗法，檢測上述箱魨魚、翻車魚 (曼波魚)、鯡魚和臭都魚之毒性，具有生物毒性者，進一步精製純化，以 LC-MS/MS 方法測定菟葵毒。第 2 年採集台灣產菟葵，同時進行季節性之生物毒性試驗，及精製純化鑑定毒成分。第 3 年在研究室飼養菟葵，探討上述魚種與菟葵共存下，檢測上述魚種蓄積菟葵毒之可行性，以建立有毒魚類會有菟葵毒之生物蓄積資訊，並提供台灣管控魚類之菟葵毒中毒事件發生。上述三種海洋生物毒，河魨毒和麻痺性貝毒之毒理機制為封閉體內電壓門控鈉離子腔體，已知具有減緩燙傷所造成之疼痛，減輕肌肉損傷造成之疼痛，而且其止痛效果僅限於施打部位而不會影響全身。菟葵毒之毒理機制為作用於細胞膜之 Na+-K+ ATPase 而導致細胞死亡。為知三種毒素之微量毒作用，另外，擬定這三年利用不同動物細胞 (含神經細胞、腸表皮細胞和血管表層細胞)，探討 3 種海洋生物毒在不同動物細胞之微量毒性作用，以進一步了解不同毒素對不同細胞之毒作用途徑和訊息傳遞之差異性，以提供日後醫療參考及作為利用三種毒素作為藥物之管控資訊。" "Palytoxin (PTX), tetrodotoxin (TTX) and paralytic shellfish poison (PSP) are marine toxins in Taiwan, which caused many poisoning accidents by ingesting seafoods in recent years. In this study, the toxicity of toxic coral Palytoa spp. and related species of sardine, sunfish and pufferfish which have caused food poisoning cases will be investigated particularly. The related toxic species from the coast of Taiwan will be collected to analyze their toxicity and to establish the DNA data base for further comparison in first year. Furthermore, the toxicity including mouse bioassay, hemolytic and enzymatic activities will be detected with PTXs extracted from Palythoa sp. in Taiwan, and the reliable standards are supposed to make according to the biological activity. The composition and concentration of purified PTXs will be also determined by HPLC-UV and LC-MS/MS analysis in the second year. In addition, the PTX bioaccumulation will be studied from Palythoa sp. to fish by using LC-MS/MS in the third year. The toxic mechanism seasonal distribution of PTX are supposed to realize, and PTX can be purified massively for further research. Furthermore, TTX, PSP and PTX are considered to use as medicine to heal the pain recently. Hence, the toxic mechanism of low dose of three toxins on different human or animal cells are necessary to study early. In each year, the different human or animal cells such as neuron cell, intestine cell and blood cell are used to examine the toxicity of low dose of PTX, TTX and PSP. These results are useful to make sure the safety of seafood and those toxins could be applied as medicinal drug."
