Abstract
本研究為三年期長期計劃之第一 年,長程目標在界定那一種營養鹽會控制超微浮游植物與大型浮游植物間的相對優勢,並進而影響東海海域有機碳的流向及 食物鍊的結構。本年中雖曾進行初期之培 養實驗,但主要工作為測試流式細胞儀鑑 別超微浮游植物的能力與準確度,以便順利處理培養實驗的樣本。我們利用2001 年 3 月的航次收集了東海陸棚區的水樣,以 流式細胞儀計數細胞後,發現水層中每單位面積的聚球藻生物量在沿岸較低,只有 2.52×109 ~ 4.28×1010 cells m-2,而向黑潮漸增加,到了黑潮海域聚球藻細胞密度可 達 1.33 ~ 2.27×1012 cells m-2 ,數量約為沿岸區的 100 ~ 1000 倍。在以螢光顯微鏡 計數聚球藻方面,我們選了 10 個樣本和流式細胞儀的結果進行比較。發現兩種方 法所得的結果類似,直線回歸顯示兩者間的斜率接近 1 但截距並非為 0 ,顯示流式細胞儀所計數的細胞數量較螢光顯微鏡計數值略低。這個差異將由變更固定溶液與調整流式細胞儀設定等兩方面加以改善。This study is the first phase of a 3-year research project. The long term goal is to identify the critical nutrient species that controls the relative dominance between ultraphytoplankton and phytoplankton of larger sizes, and to examine how this change in dominancy affects the carbon cycling and food chain structure in the East China Sea. Although a certain amount of nutrient enrichment experiments have been performed in this year, major effort was placed on testing a newly purchased flow cytometer to evaluate its performance of identifying ultraphytoplankton and in cell counting precision. A batch of water samples was collected during a cruise to the East China Sea in March, 2001. Flow cytometer was used to enumerate the abundance of Synechococcus, a dominant ultraphytoplankton in the East China Sea. The result indicated low population densities ranged from 2.52×109 ~ 4.28×1010 cells m-2 in the coastal zone. The cell density increased gradually toward the shelf break and reached 1.33 ~ 2.27× 1012 cells m-2 in Kuroshio. This level was 100 to 1000 times higher than abundance in the coastal zone. To compare cell abundance 4 obtained by flow cytometry and microscopy, 10 selected samples were examined under a fluorescent microscope. Cell numbers obtained from these 2 methods were similar, and a regression line with a slope close to 1 was obtained. However, the intercept of the equation was significant different from zero, which indicates the cell number from flow cytometry was somewhat lower. Ingredients of the fixative used and settings on flow cytemeter will be adjusted to obtain data of better qualities.
