Description

Abstract
"本申請案為整合型計畫「全球變遷對西北太平洋臺灣海域海洋生物地球化學與生態系統 影響之長期觀測與研究(一)」中的子計畫,於去年提出後獲得國科會一年的經費支助,申請 人根據執行至今的初步成果小幅修訂計畫書,並繼續申請後續兩年的經費。由於全球暖化會 導致海洋表層溫度升高,造成層化現象,使得浮游植物生長所需的營養鹽更加缺乏,所以申 請人擬利用未來兩年繼續探討東海中的矽藻在何種環境條件下會受到營養鹽的限制。本項計 畫要測試的假說是:在東海陸棚受陸源淡水影響較小的海域中,營養鹽躍層的深度控制了矽 藻受營養鹽限制的強度,而躍層以下的氮、磷、矽組成比例則決定那一種營養鹽限制生長。 所採用的方法是參與基隆嶼季節性航次和東海南部跨陸棚航次,採取樣本後進行缺氮、缺磷、 與缺矽指標基因 mRNA表現量的測定,並將表現量和水文參數進行比較。另外針對缺磷與缺 矽指標基因測試尚未完成部分,也將在計畫中以全轉錄體定序及聚合脢連鎖反應進行補強。 本申請案與其他子計畫間有密切的合作與互動,所需的水文資料和營養鹽濃度將來自「基礎 生產力變動」子計畫,而所得營養鹽限制資料也能匯入現有的生態模式,配合其他子計畫共 同瞭解東海生態系在環境變遷時的反應。 " "This proposal is a branch project under the integrated research project “Effects of Global Change on Ocean Biogeochemistry and Ecosystems in the Seas surrounding Taiwan in the Northwest Pacific (I)” (ECOBEST-I). The first phase of this research has been funded for 1 year, and the current application represents an adjusted and improved version for continuing this research in the second and the third years. Global warming inevitably causes temperature to rise in ocean’s surface layer, which will intensify the degree of stratification in the water column and reduce the nutrient supply for phytoplankton growth. To provide correct mechanisms and information to the prediction of future changes in the East China Sea (ECS), this proposal aims to investigate the environmental conditions that stimulate diatoms to display symptoms of nutrient stress in a period of 2 years. Diatoms are emphasized because they support the grazing food chain and help the sinking of organic carbon in marine environments. In the proposal, the hypothesis to be tested is: in the part of ECS where the influence of terrestrial runoff is minor, the depth of nutracline controls the severity of nutrient stress in diatoms while nutrient stoichiometry below nutracline controls the chemical species causing stress. Oceanographic cruises will be conducted in the ECS, and water samples will be collected for the measurement of mRNA levels of indicator genes for nitrogen-, phosphorus-, and silica-stress. The measured mRNA levels will be analyzed with respect to hydrographic parameters. Two types of survey are designed with a cross-shelf survey to evaluate spatial variations and a single-station seasonal survey to evaluate seasonal variations. In addition, to complete the tests related to phosphorus-, and silica-stress genes, transcriptome sequencing and polymerase chain reaction will be performed on algal cultures and natural assemblages of diatoms. This proposal will work collaboratively with other branch projects under ECOBEST-I. Hydrographic information and nutrient concentrations will be provided by the “variations in primary productivity” branch project. The mechanisms of nutrient stress obtained by this proposal will also be fed to an existing ecological model of ECS in order to achieve a comprehensive understanding of responses in the ECS under the influences of environmental changes."
