Description

Abstract
This three-year research proposal will use flow cytometry combined with meta-omics strategies to investigate the relationship between picoplankton succession and environmental factors in the subtropical Kuroshio Current. The community structures and their abundances of picoplankton are important parameters for marine biogeochemistry. Furthermore, because of their susceptibility to environmental changes, the assemblage of picoplankton might be an indicator of environmental change. Based on the results of the previous research projects supported by the Ministry of Science and Technology (Taiwan), in addition to the long-term observation of the spatial and temporal distribution of picoplankton in the Kuroshio waters, it will focus on the application of meta-omics strategies (i.e., small subunit rRNA gene phylogeny and meta-transcriptomic analysis) to analyze the community structure and the physiological status of picoplankton. These results will be integrated with various biogeochemical parameters measured by the collaborative research teams. Establishing complete observational data on the community composition and the spatial-temporal distribution of picoplankton and the in situ gene expression profiles in the subtropical Kuroshio waters will help deduce the possible impacts of global warming on the plankton ecology and the biogeochemical cycles in the northwest Pacific. 本研究申請案計畫以流式細胞儀結合環境基因體學相關分析技術,針對亞熱帶黑潮流域超微浮游生物族群演替與環境因子之關係進行探討。超微浮游生物各物種的族群結構及數量不僅為海洋生地化相關研究的重要參數;其對於環境變化的易敏感性,使得超微浮游生物的族群消長也可做為環境變遷的指標。本申請案將奠基於之前科技部專題研究計畫成果,預計以3年期間,除了持續針對亞熱帶黑潮流域超微浮游生物時空分布進行長期觀測外,將更進一步著重於應用全轉錄基因體學相關技術分析該水域超微浮游生物的生理狀態,以評估環境因子改變對於超微浮游生物生態的影響程度。這些結果將與協同研究團隊測得的各式生地化參數進行整合,以期能對亞熱帶黑潮海域超微浮游生物族群結構與時空分布建立完整的觀測資料,並藉以推論全球暖化對於西北太平洋超微浮游生物生態與生地化循環可能造成之影響。
