Description

Abstract
養殖吳郭魚的加工產業中,特別是台灣鯛冷凍魚片的加工,常伴隨而來大量的加工共/副產(每 年約4500 公噸),如魚皮、魚頭及魚骨,一直是低度利用之加工素材;然而這些原料富含大量的蛋 白質,魚鱗、魚皮中的膠原蛋白、及附著於魚頭及魚骨的肌纖維蛋白等。本實驗室於 J. Functional Food (2015) 發表研究成果指出,吳郭魚魚皮膠原蛋白為多種活性胜肽之前驅物,活性胜肽的氨基 酸序列的覆蓋率占總序列的89.7%,特別是抑制血管收縮轉換酵素活性(RL、IR…)、抗健忘活性 (GPGG、PGP、PG、GP)、抗血栓活性(GPRG、GPR…)及抗氧化活性(AY、EL、WY、PHD、KD…) 胜肽等,因此本計畫將著重於吳郭魚皮中膠原蛋白水解產物之活性胜肽之分離純化與其多重功能性 驗證,先以in vitro 實驗進行活性評估後,再進行in vivo 動物試驗,以原發性高血壓大鼠及小鼠為 動物實驗對象,各別探討活性胜肽(抑制ACE 活性)之降低大鼠血壓功效,及以小鼠模式的水迷宮 試驗,建立抗建忘胜肽功能性與小鼠記憶學習模式之相關性,進而提供開發吳郭魚皮膠原蛋白水解 產物(多功活性胜肽)具保健功效產品之參考。 In Taiwan, industrial processing of cultivated tilapia especially the processing of produced massive (~4.5 k tons/per year) co-/by-products of fish scales and skin. Generally, these co-/by-products are underutilized processing materials which are relatively rich in proteins such as collagens from fish skin, and muscle proteins from residual flesh that are attached to fish bones and heads. According to our recent study published in Journal of Functional Foods, tilapias collagens contain precursors of multiple bioactive peptides (89.7% sequence coverage) in particular ACE inhibitor (RL、IR…), antiamnesic (GPGG、PGP、PG、GP), antithrombotic (GPRG、GPR…) and antioxidant (AY、EL、WY、PHD、KD…) activities. Consequently, this project focuses on the separation and purification of bioactive peptides from tilapias collagen hydrolysates, and assessment of their multi-functional activities. Firstly, the experiments undergo in vitro assessments on various activities. Following the in vitro results, in vivo experiments will be performed in order to evaluate the inhibition effects of purified bioactive peptides on hypotension effects using an animal model with spontaneous hypertension rats (SHR). In the meanwhile, the antiamnesic activities of purified bioactive peptides will be investigated using a Morris water maze system with small rats. The projected experimental results can be used as a reference for developing nutraceutical products (multi-functional bioactive peptides) with preventing relevant diseases using skin collagen hydrolysates of tilapia processing co-/by-products.
