分別純化九株七種不同的弧形菌的遺傳體DNA,然後利用BclⅠ的限制酶及三種同功限制酶(Sau3AⅠ,MboⅠ及DpnⅠ)來研究GATC核苷酸序列中腺嘌呤鹽基是否有甲基化的現象。從這次的實驗中,我們發現所有被研究的海洋病原性弧形菌其腺嘌呤都有甲基化的現象,也就是在GATC序列中的腺嘌呤鹽基都以6-甲基-腺嘌呤的形式存在。但是這樣的機制是否與大腸菌相同以及是否有其特殊的生物特性却有待研究。Genomic DNA obtained from nine strains categorized of seven species ofmarine pathogenic Vibrio species were examined to determine whether themethylation of adenine residue occurred at GATC sequence.Several restrictionendonucleases,such as Sau3AⅠ,MboⅠ,DpnⅠ and BclⅠ,were used.In thepresent communication,we demonstrated that all pathogenic vibrios testedwere Dam phenotype,which presented 6-methyladenine at GATC sequence.The mechanism and the biological significance of adenine methylation occurredin marine pathogenic vibrios remain to be studied.
