Through the fry release, the amount of natural population resources can be increased, which is currently recognized as a management measure that can effectively enhance fishery resources and drive marine economic growth and food production. In order to avoid the narrowing of the genetics of inbreeding of released fish fry and the destruction of the genetic structure of the coastal wild populations around Taiwan. The purpose of this work was to analyze the genetic diversity of the wild and farmed yellowfin seabream using microsatellite markers and ultimately propose a standard operation procedure for the genetic analysis of yellowfin seabream. The results showed that a total of 133 alleles could be generated by using the four microsatellite DNA loci in the yellowfin seabream species, and the H_o, H_e and PIC data representing genetic diversity were all above 0.5. According to the analysis results, both the wild and release populations of yellowfin seabream showed highly genetic polymorphisms, and they may be in the state of undergone hybrid breeding. Through this research of the yellowfin seabream, a genetic database was established. In the future, when evaluating the release fry of yellowfin seabream, the database can provide a reference basis for the genetic management and fry release of farmed fish, and establish molecular markers to assist in the selection and breeding of suitable fish for production and release. The platform can reduce the problem of gene narrowing caused by fry release, effectively assists in the cultivation of fisheries, and ensures that the genetic diversity of gene pools in the wild population is not affected by the fry release.en_US
